چکیده
Clerodendrum cyrtophyllum Turcz has been used in traditional medicine for the treatment of various diseases. In spite its therapeutic applications, research on toxicity and teratogenicity is still limited. The study aimed to investigate developmental ethanol extract C. (EE) zebrafish embryo model. Major compounds from crude Clerodendron leaves were determined using HPLC-DAD-Orbitrap-MS analysis. EE investigated Zebrafish embryos at 6 h post-fertilization (hpf) treated with different concentrations. Egg coagulation, mortality, hatching, yolk sac edema, pericardial edema recorded each day during a 5-day exposure. At time point 120 hpf, body length, area, heartbeat area assessed. order elucidate molecular mechanisms EE, we further evaluated effects expression genes involved signaling pathways affecting fish embryo’s development such as heart (gata5, myl7, myh6, has2, hand2, nkx 2.5), oxidative stress (cat, sod1, gpx4, gstp2), wnt pathway (?-catenin, wnt3a, wnt5, wnt8a, wnt11), or cell apoptosis (p53, bax, bcl2, casp3, casp8, casp9, apaf-1, gadd45bb) qRT-PCR Our results demonstrated that three major components including acteoside, cirsilineol cirsilineol-4'-O-?-D-glucopyranoside identified EE. exposure 6–96 doses ranging 80 200 ?g/mL increased mortality reduced hatching rate. 20 40 until 72–120 hpf induced series malformations, spine deformation, shorter length. Based two prediction models teratogenic index (TI), 25% lethality concentration (LD25) no observed-adverse-effect level (NOAEL), considered TI (LC50/EC50) LD25/NOAEC values 96 reaching 3.87 15.73 respectively. mRNA levels p53, bax/bcl2, gstp2, nkx2.5, wnt11, gadd45bb gata5 significantly upregulated by while hand2 bcl2 downregulated. These provide evidence stages an insight into potential embryonic development.

Optimization of Ultrasonic-Assisted Extraction and Radical-Scavenging Capacity of Phenols and Flavonoids from Clerodendrum cyrtophyllum Turcz Leaves
Ultrasonic-assisted extraction (UAE) was developed to extract phenolic and flavonoid antioxidants from Clerodendrum cyrtophyllum Turcz leaves. The optimal experimental parameters for antioxidant extraction from C. cyrtophyllum leaves were measured using single-factor experimentation combined with response surface methodology (RSM). Total phenolic content (TPC) and total flavonoid content (TFC) ...
متن کاملEmbryo and Developmental Toxicity of Cefazolin Sodium Impurities in Zebrafish
Cefazolin sodium is an essential drug that is widely used in clinical therapy for certain infective diseases caused by bacteria. As drug impurities are considered to be one of the most important causes of drug safety issues, we studied embryotoxicity, cardiotoxicity, and neurotoxicity of nine cefazolin sodium impurities in zebrafish embryo and larvae for the objective control of impurity profil...
متن کاملAutomated zebrafish chorion removal and single embryo placement: optimizing throughput of zebrafish developmental toxicity screens.
The potential of the developing zebrafish model for toxicology and drug discovery is limited by inefficient approaches to manipulating and chemically exposing zebrafish embryos-namely, manual placement of embryos into 96- or 384-well plates and exposure of embryos while still in the chorion, a barrier of poorly characterized permeability enclosing the developing embryo. We report the automated ...
متن کاملDevelopmental toxicity of the dithiocarbamate pesticide sodium metam in zebrafish.
Sodium metam (NaM), a dithiocarbamate, is a general agricultural biocide applied prior to planting for the elimination of nematodes, soil pathogens, and weeds. There is a remarkable paucity of information about the mechanism of action and the risk that dithiocarbamates may pose to developing vertebrates. We have characterized NaM toxicity during early life stage exposure in zebrafish. Zebrafish...
متن کاملExogenous Nucleotides Antagonize the Developmental Toxicity of Ethanol In Vitro
The objective of this study was to assess whether nucleotides supplementation in vitro could suppress ethanol-induced developmental toxicity in mouse. The models of whole embryo culture (WEC) and midbrain (MB) cell micromass culture were used in this study. In WEC system, exposure to 4.0 mg/mL ethanol for 48 h yielded various developmental malformations of the mice embryos. Nucleotides suppleme...
